Recombinant DNA is a chapter where the order is the answer. Swap two steps, or call DNA ligase a "restriction enzyme", and a short-answer question goes wrong. An AI tutor is a fast way to get a first draft of the sequence at 11 pm. But the draft is only useful once you have checked every step and every name against the book your exam is set from: NCERT Class 12 Biology, Chapter 9, Biotechnology: Principles and Processes.
Here is the order NCERT gives, a prompt to use, and a checklist for marking the AI's answer.
What are the steps of recombinant DNA technology in NCERT?
Section 9.3 of the NCERT chapter lists the processes "in specific sequence". Here they are with the tool the chapter names for each:
Isolation of DNA: break cells open with lysozyme (bacteria), cellulase (plant cells) or chitinase (fungus), remove RNA with ribonuclease and proteins with protease, then precipitate the DNA with chilled ethanol.
Cutting DNA at specific locations: restriction endonucleases. Cut the source DNA and the vector with the same enzyme.
Isolating the fragment you want: agarose gel electrophoresis, staining with ethidium bromide under UV light, then cutting out the band (elution).
Ligation into a vector: DNA ligase joins the gene of interest to a cloning vector such as the plasmid pBR322.
Transfer into the host: make E. coli "competent" with a divalent cation such as calcium, then heat shock at 42°C.
Culturing the host at large scale: bioreactors, where 100 to 1000 litres of culture can be processed.
Extracting the product: downstream processing, which means separation and purification, then formulation and quality control.
PCR (section 9.3.3) can sit between steps 3 and 4 if you need more copies of the gene. Figure 9.6 shows 30 cycles amplifying the region about 1 billion times, using Taq polymerase from Thermus aquaticus, per the same chapter.
What should you ask an AI tutor about recombinant DNA?
Do not ask "explain recombinant DNA". You will get a smooth essay that hides the mistakes. Ask for something you can mark. Copy this:
"List the steps of making recombinant DNA in order, as in NCERT Class 12 Biology Chapter 9. Number them."
"For each step, name the enzyme, tool or method used, and say in one line why that step must come before the next one."
"Now quiz me: shuffle the steps and ask me to put them back in order."
The "why before the next one" line is where the learning is. You cannot ligate before you cut, and you cannot select transformants before the plasmid carries a selectable marker. If the AI cannot justify an order, that step needs checking.
How do you check the AI's answer against NCERT?
These are easy places for an answer to drift from the textbook. Check each one against the NCERT PDF:
Endonuclease, not exonuclease. NCERT says exonucleases remove nucleotides from the ends, while endonucleases cut at specific positions within the DNA.
The first restriction endonuclease NCERT names is Hind II, which recognises a specific sequence of six base pairs. Hind III appears later as a site on pBR322; do not mix them up.
EcoRI's cut. EcoRI cuts between G and A only when the palindrome GAATTC is present, leaving sticky ends.
Blue-white selection. With a chromogenic substrate, colonies whose plasmid has no insert turn blue; recombinant colonies, where the β-galactosidase gene is inactivated by the insert, give no colour. Check that the answer has not flipped this.
pBR322 and antibiotics. Insert at the BamH I site and the plasmid loses tetracycline resistance. Recombinants grow on ampicillin but not on tetracycline.
Spellings and symbols. NCERT spells it Agrobacterium tumifaciens, with the Ti plasmid; most other sources write tumefaciens. If the AI spells a name differently from the book, ask your teacher which version your board accepts.
If two names still blur together, the Fermi Concept Library gives each biology concept its own lesson, so you can read up on vectors or restriction enzymes separately before going back to the sequence.
What if the AI gets the order wrong?
Tell the tutor exactly where it disagrees with NCERT and ask it to explain why the book's order works. Arguing with a wrong answer makes you use the textbook as evidence, which is the skill an exam question tests. We have also written about what to say when an AI tutor gives your child a wrong answer, which works just as well for students.
To practise the cutting step with numbers, try counting the DNA fragments a restriction enzyme makes. For the story behind the first experiments, read how Paul Berg created recombinant DNA.
Fermi's AI tutor teaches rather than just answers: it notices where you are stuck, decides whether to explain, ask or show, and checks your understanding as it goes. Keep the NCERT chapter open beside it, and let the book have the final word.
